Pertahanan Kota Brest merupakan pertempuran pertama dalam Operasi Barbarossa, iaitu pencerobohan Kuasa Paksi terhadap Kesatuan Soviet yang dilancarkan pada 22 Jun 1941. Tentera Darat Jerman menyerang tanpa amaran, dan menjangkakan dapat menawan Brest pada hari pertama hanya dengan menggunakan infantri dan artileri. Namun begitu, mereka hanya berjaya menawan kubu itu selepas seminggu, dan selepas dua kali pengeboman oleh Luftwaffe. Ramai anggota pertahanan telah terbunuh atau ditawan.
